چکیده

Improved electron detectors and image-processing techniques will allow the structures of macromolecules to be determined from tens of thousands of single-particle cryo-EM images, rather than the hundreds of thousands needed previously.

High Resolution Imaging of Radiation Sensitive Biological Samples

Cryo-electron microscopy has seen incredible progress recently. From the first reconstruction of frozen hydrated samples to recent atomicresolution structures, structural biology is undergoing a revolution. However, until recently, the resolution of cryo-EM structures was often significantly lower than that of X-ray crystallography. We have reached a point where, for an increasing number of sam...

Cryogenic-temperature electron microscopy direct imaging of carbon nanotubes and graphene solutions in superacids.

Cryogenic electron microscopy (cryo-EM) is a powerful tool for imaging liquid and semiliquid systems. While cryogenic transmission electron microscopy (cryo-TEM) is a standard technique in many fields, cryogenic scanning electron microscopy (cryo-SEM) is still not that widely used and is far less developed. The vast majority of systems under investigation by cryo-EM involve either water or orga...

Beam-induced motion correction for sub-megadalton cryo-EM particles

In electron cryo-microscopy (cryo-EM), the electron beam that is used for imaging also causes the sample to move. This motion blurs the images and limits the resolution attainable by single-particle analysis. In a previous Research article (Bai et al., 2013) we showed that correcting for this motion by processing movies from fast direct-electron detectors allowed structure determination to near...

A new approach for the direct visualization of the membrane cytoskeleton in cryo-electron microscopy: a comparative study with freeze-etching electron microscopy.

An improved unroofing method consisting of tearing off the cell membrane using an adhesive electron microscopy (EM) grid instead of vitreous ice sectioning (cryo-sectioning) has enabled us to panoramically view the membrane cytoskeleton in its native state with extremely high contrast. Grids pre-treated with Alcian blue were placed on cells, and a portion of the dorsal plasma membrane was trans...

Adaptive nonparametric detection in cryo-electron microscopy

Cryo-electron microscopy (cryo-EM) is an emerging experimental method to characterize the structure of large biomolecular assemblies. Single particle cryo-EM records 2D images (so-called micrographs) of projections of the three-dimensional particle, which need to be processed to obtain the threedimensional reconstruction. A crucial step in the reconstruction process is particle picking which in...
